Maize growth response to 2 productsWhether you are from a research lab, or from a commercial branch, you should know about the quality of the product(s) you use. In the example at the right, one product resulted in a strong increase in fresh weight of maize, whereas the other one had no effect at all. However, the growth effect shown was not mediated by AM fungi, but by other effects. Studying products by bioassays gives us indication about the activity of AM fungi, but also about nutrient (fertilizer) content of products.

Note for your application that early plant responses may be dose-dependent; products with low concentration of AM fungi are not expected to show any AM fungal mediated effects after short times of 2-3 weeks, as AM fungi need some time to establish symbiosis, propagate, and promote plant growth.

We have reduced our portfolio of AM fungal inocula to the fungus that is used in vast majority of research approaches.

We offer research-grade quality AM fungal inoculum based mainly on fungal spores as propagules, a low number of root fragments may also be present. If you need material for larger scale research attempts (e.g. for field experiments), please inquire whether more economical but less well defined bulk standard inoculum quality is available. All material is produced aseptically, in vitro.  We supply AMF propagules in fine clay-based (calcined attapulgit) or diatom earth ('diatomite') powder. Specific formulations like in liquid or in gel may be produced and delivered on demand, but this may take several months.

The material is identical to the carrier in the powder based products, but contians no water (apply half of the inoculum (50% water content) weight for control experiments). It can, e.g., be use for control experiments or also for dilution of inoculum. Only sold in combination with SYMPLANTA inoculum. Material is gamma-irradiation sterilized.

Services

Species monitoring - NGS (MiSeq) based

We adopted our originally 454-based system to the MiSeq platform and sucessfully conducted a number projects since June 2017.  
Please inquire well in advance for any related projects, as we may need about 3 months for individual projects. For new customers we may request a 50% pre-payment.

DNA based high throughput species identification 'MiSeq' from various (field roots, culture substrate, etc.) samples.
PCR in triplicate, pooling, 450-500 bp based identification of AM fungi at species level, using a well established marker in the LSU rRNA gene region.
AMFMiSeq, price: 9600,- for an average project of 96 samples (100,- € per sample);
please note that costs range from 80-200 € per sample, dependent on sample number (small numbers may be expensive) and type (DNA or roots, etc.), please inquire!

Species identification - DNA based

DNA based species identification 'basic' from individual spores or roots (material provides must be from an isolate [single species]).
Identification of AM fungi based on PCR product (amplicon) sequencing, one sequence of 450-500 bp of the D2 region of the LSU rRNA gene, consensus including nucleotide polymorphisms. This fragment usually is sufficient for species identification using our database (provided the species is characterized). A maximum likelyhood phylogenetic analysis and a phylogenetic tree of the respective genus including the query sequence is included.

Inoculum viability and efficiency tests

Bioassay for quantification of the % root length colonization (RLC, in %) efficiency of substrates, products (e.g., commercial), or soil.
Based on a standard procedure (inoculum application in 3-4 dilutions, 5 replicates each, root staining, % root length colonization analyses 5-7 weeks after inoculation) using a selected lettuce variety as test plant. If you need testing of other target plants, please inquire. Note that will need about 8 weeks for the entire procedure.

Inoculum viability and efficiency tests

Standardised test of the % of viable spores in substrates, products (e.g., commercial).
Based on a standard procedure (MTT staining, 4 replicates, estimating number of putatively viable spore content). Note: this is only applicable with suited substrates (microscopic counting has to be possible) and also suited AMF species. Also note, that MTT positive reaction is not in all cases a 100% proof vor viability - if you need to be sure about colonization potential, you should apply a bioassay. MTT can be efficiently used for process control and QC of defined formulations.

Species identification - DNA based

DNA based species identification 'detailed', from individual spores or mixed material (spores, roots, soil, ...).
PCR, cloning, RFLP screening, and DNA sequencing based identification of AM fungi, including 10 diverse >1500 bp sequences.
Inquire for complex samples, which e.g. demand multiple initial PCR reactions (pooled before cloning), and screening of larger clone numbers (approx. additional costs 100 €, but depends on number of sequences to be provided).

Species identification - spore morphology based

AM fungi surveys from soil; spore morphology based. Analysis of one normal sample (up to 5 AM fungal species in difficult substrate; up to 8 AM fungal species in a easy to handle substrate; if your samples fall in the high complexity category, we will inform you before any analyses).
AM fungal spore abundance is determined by spore extraction from 50 ml soil, taxonomic identification, and counting of the different morphotypes.
Includes photographic documentation and vouchers.
